Diese Masterarbeit beinhaltet die rekombinante Produktion einer aktiven Sojaprotein-abbauenden Protease in Escherichia coli, welche als Futtermittelzusatz eingesetzt werden kann. Für diesen Zweck wurde ein Serinprotease-kodierender DNA Strang aus Xanthomonas glycines synthetisiert. Das Gen wurde in den pET26b(+) Vektor ligiert und in dieser anschließend in den E. coli Expressionsstamm BL21(DE3) transformiert. Nach einer 200 mL Fermentation wurden die Zellen mittels Ultraschall aufgeschlossen um an die intrazellulären Proteine zu gelangen. Da die Proteine Inclusion Bodies bildeten, erfolgte im Anschluss ein Refolding mit darauffolgender Dialyse. Zum Schluss wurden die Proteine mittels IMAC aufgereinigt und aufkonzentriert. Die Charakterisierung der Proteine erfolgte mit Hilfe von Acrylamid-Gelen und Western Blots. Die Konzentration wurde mit Hilfe eines Densitometers und eines Bicinchoninsäure Assays ermittelt. Die Aktivitätsmessungen erfolgten mittels verschiedenen Substraten: Azocasein, p-Nitrophenyl 2-(trimethylsilyl) ethyl Carbonat und zwei unterschiedliche Nitroanilide. Die Sequenzierung des Klonierungsvektors mit dem Protease-Gen als Insert zeigte keine Fehler in der DNA Abfolge. Die Identifizierung mittels Acrylamid-Gel und Western Blot zeigte eine Bande mit dem erwarteten Molekulargewicht. Die berechnete Konzentration von 3 mL aufgereinigtem Endprodukt betrug etwa 200 ppm. Abgesehen von p-Nitrophenyl 2-(trimethylsilyl)ethyl Carbonat zeigten alle weiteren Substrate nur eine sehr geringe Aktivität der hergestellten Protease. Daher muss entweder der Downstream-Prozess optimiert werden oder andere Protease-kodierende Gene verwendet werden.*****This master thesis deals with the recombinant production of an active soy protein digesting protease in Escherichia coli, which can be used as feed additive. Therefore, a serine protease encoding DNA strain, obtained from Xanthomonas glycines was ligated into a pET26b(+) vector which was transformed into the E. coli expression host BL21(DE3). After fermentation with a total culture volume of 200 mL cells were disrupted through sonication to get the intracellular protein. As the proteins formed inclusion bodies, refolding and subsequent dialysis was required. Finally, the proteins were purified and concentrated with an IMAC system. To characterize the protein, acrylamide gels and Western blots were performed. The concentration of the produced protein was determined with a densitometer and a bicinchoninic acid assay. For activity measurements, azocasein, p-nitrophenyl 2-(trimethylsilyl) ethyl carbonate and two different nitroanilides were used as substrates. Sequencing of the cloning vector with the protease gene as insert before transforming it into the expression host did not show any mistakes in the DNA. The identifications with the acrylamide gel and western blot showed a band at the expected molecular weight. The calculated concentration of 3 mL of the final purified protein product was approximately 200 ppm. Apart from p-nitrophenyl 2-(trimethylsilyl) ethyl carbonate, the other activity assays showed only little activity of the produced protease. Therefore, it is either necessary to optimize the protein downstream processing or to focus on other DNA genes which encode different proteases.
